3M Petrifilm Interpretation Guide 3M Petrifilm Coliform Count Plates This guide familiarizes you with results on 3M Petrifilm Coliform Count Plates (CC). For further information, please contact the 3M Mic robiology representative nearest you. Petrifilm Coliform Count plates contain modified Violet Red Bile (VRB) nutrients, a cold-water soluble gelling agent and a tetrazolium indicator that facilitates colony enumeration. The top film traps gas produced by the lactose fermenting coliforms. Time and Temperature of incubation as well as interpretation of the Petrifilm Coliform Count plates vary by method. I S O defines coliforms by their ability to grow in method-specific, selective media. ISO method 4832, enumerating coliforms by the colony count technique, defines coliforms by colony size and acid production on VRB with lactose (VRBL) agar. On Petrifilm Coliform Count plates, these acid-producing coliforms are indicated by red colonies with or without gas (see circle ). ISO method 483, enumerating coliforms by the most probable number (MPN) method, defines coliforms by their ability to grow and produce gas from lactose in a selective broth. On Petrifilm Coliform Count plates these coliforms are indicated by red colonies associated with gas (see circle 2). AOAC INTERNATIONAL and FDA (Food and Drug Administration)/BAM define coliforms as gram-negative rods which produce acid and gas from lactose during metabolic fermentation. Coliform colonies growing on the Petrifilm Coliform Count plate produce acid which causes the ph indicator to deepen the gel colour. Gas trapped around red colonies indicates coliforms (see circle 2).. The Petrifilm CC plate is suitable to use for detection of both total coliform and thermotolerant (faecal) coliform. AOAC, AFNOR and NMKL have validated the use of Petrifilm CC plates under specific conditions. Interpretations of 3M Petrifilm Coliform Count Plates according to protocols described by the following bodies: AOAC, NORDVAL and AFNOR a. Reading following AOAC Official Methods SM for total coliforms (986.33, 989.0 and 99.4) Incubation: Enumeration of coliforms in milk, raw milk and dairy products (Official Methods 986.33 and 989.0): incubate 24h +/- 2h at 32 C +/- C. Enumeration of coliforms in all foods, except those listed above (Official Method 99.4): incubate 24h +/- 2h at 35 C +/- C. Interpretation: Coliforms: Count all red colonies with gas. 65 coliforms, AOAC Official Methods SM b. Reading following NORDVAL validated method for total coliform (NORDVAL certificate n 3) Incubation: 24h +/- 2h at 37 C +/- C. Interpretation: Coliforms: Count all red colonies with gas. 67 coliforms, NMKL method. NORDVAL validated method.

Reading following AFNOR approval for total coliforms (certificates number 3M 0/2-09/89A and 3M 0/2-09/89B) Incubation: 24h +/- 2h at 30 C +/- C Interpretation: Comparison to ISO 4832 method (certificate 3M 0/2-09/89A): Count all red colonies with or without gas. Comparison to ISO 483 method (certificate 3M 0/2-09/89B): Count only red colonies with gas. 97 coliforms, AFNOR approved method compared to ISO 4832 method. 72 gas producing coliforms, AFNOR approved method compared to ISO 483 method. d. Reading following AFNOR approval for thermotolerant coliforms (certificate number 3M 0/2-09/89C) Incubation: 24h +/- 2h at 44 C +/- C Interpretation: Comparison to NF V08-07 method: Count all red colonies with or without gas. 2 coliforms, AFNOR approved method compared to NF V08-07 method.

3M Petrifilm Coliform Count Plates As the coliform count increases, the gel colour deepens, as shown in figures 2 through 6. 2. 3. 3 2 No growth Colony count = 0 Background bubbles are a characteristic of the gel and are not a result of coliform growth. Background gas bubbles are small to pin-point in size and do not have a colony associated with them. Gas producing colonies count: 8 Non-gas producing colonies count: 7 Total count: 5 Figure 3 shows how bubble patterns may vary. Sometimes gas disrupts the colony so that the colony outlines the bubble (see circles and 2). Gas bubbles are usually within approximately one colony diameter. Artifact bubbles may result from improper inoculation of the Petrifilm CC plate or from trapped air within the sample. They are irregularly shaped and are not associated with a colony (see circle 3). 4. 5. Gas producing colonies count: 83 Non-gas producing colonies count: 29 Total count: 2 The preferable counting range (total colony population) on Petrifilm CC plates is 5-50 colonies. Do not count colonies that appear on the foam dam since they are removed from the selective influence of the medium (see circle ). Estimated total count: 30 The Petrifilm CC plate circular growth area is approximately 20 cm 2. Estimates can be made on plates containing greater than 50 colonies by counting the number of colonies in one or more representative squares and determining the average number per square. Multiply the average number by 20 to determine the estimated count per Petrifilm CC plate. Further dilution of the sample is recommended to determine an accurate count.

TNTC (Too Numerous To Count) plates Petrifilm CC plates with colonies that are TNTC have one or more of the following characteristics: many small colonies, many gas bubbles, and a deepening of the gel colour. Gas producing colonies count: 4 When high numbers of non-coliforms organisms such as Pseudomonas are present on Petrifilm CC plates, the gel may turn yellow. Gas producing colonies count: 2 Bubbles 8. 9. 2 3 4 5 6 7 8 9 Food particles are irregularly shaped and are not associated with gas bubbles (see circle ). Here are additional examples of various bubble patterns associated with a colony. All of them should be enumerated as coliform.

Reminders for use Storage Store unopened packages frozen or refrigerated at 8 C ( 46 F). Use before expiration date on package. In areas of high humidity where condensate may be an issue, it is best to allow packages to reach room temperature before opening. 2 To seal opened package, fold end over and secure with tape or a clip. 3 Keep resealed package at 25 C ( 77 F) and 50% RH. Do not refrigerate opened packages. Use Petrifilm plates within one month after opening. Sample Preparation Weigh or pipette food product into an appropriate sterile container such as stomacher bag, dilution bottle, Whirl-Pak bag, or other sterile container. If necessary, use appropriate sterile diluents: 4 5 6 peptone salt diluent (ISO method 6887) (Maximum Recovery Diluent), Butterfield s phosphate buffer (IDF phosphate buffer, K2HPO4 at 0.0425 g/l, adjusted to ph 7.2), 0.% peptone water, buffered peptone water (ISO method 6579), saline solution (0.85-0.90% weight to volume), bisulphite-free letheen broth or distilled water. Blend or homogenize sample as per current procedure. Adjust ph of the diluted sample between 6.6 and 7.2. for acid products, use N NaOH, for alkaline products, use N HCl. Do not use buffers containing citrate, bisulphite or thiosulphate, they can inhibit growth.

Inoculation 7 8 9 Place Petrifilm count plate on level surface. Lift top film. With pipette perpendicular to Petrifilm count plate, place ml of sample onto centre of bottom film. Carefully roll top film down to avoid trapping air bubbles. Do not let top film drop. With flat side down, place spreader on top film over inoculum. Gently apply pressure on spreader to distribute sample over circular area before gel is formed. Do not twist or slide the spreader. Lift spreader. Wait at least one minute for gel to solidify. Incubation Interpretation <70 F 0 Incubate count plates with clear side up in stacks up to 20. Incubation time and temperature vary by method. 2 Petrifilm count plates can be counted on a standard colony counter or other illuminated magnifier. Refer to the Interpretation Guide section when reading results. Colonies may be isolated for further identification. Lift top film and pick the colony from the gel. Most common approved methods: Total coliforms AOAC Official Methods 986.33 and 989.0 (milk, raw milk, other dairy products): Incubate 24h ± 2h at 32 C ± C. AOAC Official Methods 99.4 (all foods): Incubate 24h ± 2h at 35 C ± C. NORDVAL validated method n 3: Incubate 24h ± 2h at 37 C ± C. AFNOR validated methods 3M 0/2-09/89A and B: Incubate 24h ± 2h at 30 C ± C. Thermotolerant (faecal) coliforms AFNOR validated method 3M 0/2-09/89C: Incubate 24h ± 2h at 44 C ± C. Incubator humidification is required at this elevated temperature.
